Developing a highly targeted and effective cell therapy requires detailed understanding of the crosstalk between cancer cells and engineered immune cells already at the pre-clinical stage. Pioneering high-value applications of functional phenomics, evorion developed the CellCity System. This innovative microfluidic platform is the first of its kind, allowing researchers to link the functional phenotype of individual cells in an array with their underlying genetic and transcriptional landscape and dynamic secretion profiles. Those connections reveal novel insights into the cellular and molecular mechanisms of cancer-immune cell interactions.

Functional Phenomics

Current multi-omic technologies generate vast amounts of data on biomedically relevant genes and proteins at single-cell resolution. However, these analyses are limited to single-timepoint measurements. Lacking detailed knowledge about localization, interactions, and activation state in living cells, the functional profiles of identified proteins and genes remain incomplete. To bridge this gap, evorion’s innovative CellCity System enables encapsulating thousands of cancer and immune cell pairs in hydrogel beads to co-culture them in a physiologically relevant 3D microenvironment for long periods of time. Via live-cell imaging, researchers can investigate functional phenotypes over time (e.g., cancer cell apoptosis, tumour growth) and correlate these observations with dynamic secretion profiles of individual single cells. Furthermore, these functional phenotypes can be linked with multi-omics data by performing single-cell transcriptional analysis after retrieving the cell-laden beads from the chip. Thus, functional phenomics enables comprehensive insights into the secretion profile and transcriptome analysis of every single cell, all in one place.
